Dr Graham Mitchell is Head of the Malaria Laboratory at Guy’s, King’s and St. Thomas’s School of Medicine. In this consortium he is responsible for the preparation of cultured parasites and their exploitation in localisation experiments aimed at improving our understanding of MSP-1 and AMA1, together with other antigens studied by the consortium as the need arises. These localisation studies employ fluorescence imaging by microscopy with CCD-driven computer collection and analysis and electron microscopy, where the techniques of routine immuno-electron microscopy and serial sectioning with 3-D reconstruction involving immuno-staining will be combined. A major target of this work is the localisation of the secondary processing site of MSP1-19.

Dr Mitchell works closely with Prof Lawrie Bannister (Centre for Neuroscience, GKT) who will be taking a day to day part in scientific activities. Other members of the Malaria Laboratory at Guy’s will also contribute to the work of the EUROMALVAC Consortium. Notably Dr Anton Dluzewski will be responsible for the immuno-electron microscopy of invading merozoites, and Dr Gabi Margos for fluorescence imaging studies. Their work, especially with regard to parasite cultivation, is supported by Mr Ian Williams. The Malaria Lab’s expert electron microscopist is Mr John Hopkins. His outstanding ability is in serial sectioning, where he routinely succeeds in compiling 70+ sections from a blood stage parasite, so allowing 3D parasite-reconstruction.
